Medicure presented the design, rationale and initial baseline data of the phase‑3 MEND‑PNPO study of the oral P5P formulation MC‑1 for treatment of the rare PNPO deficiency at the 16th European Epilepsy Congress. The open‑label, multicenter 52‑week study began in January 2023 and targets about 10 genetically confirmed patients aged two years and older; so far nine patients have been enrolled and three have completed the one‑year follow‑up. Starting doses ranged from 14 to 58 mg/kg/day, measured steady‑state P5P levels were 59.8–165 ng/ml, and MC‑1 is not yet approved by the FDA.
