The FDA has approved Ziihera for first-line treatment of adults with inoperable, locally advanced or metastatic HER2-positive gastroesophageal adenocarcinoma. Two combination therapies with Ziihera were approved, including one with the PD-1 inhibitor tislelizumab; both reduced the risk of progression or death by 35% in the phase 3 trial and extended median progression-free survival to 12.4 months versus 8.1 months. The Ziihera combination lowered the risk of death by 28% in the overall HER2-positive population and achieved a median overall survival of 26.4 months versus 19.2 months with trastuzumab plus chemotherapy.
