ORIC Pharmaceuticals has launched the global phase‑3 Himalayas‑1 study for patients with metastatic castration‑resistant prostate cancer after treatment with Abiraterone. About 600 patients at more than 250 sites in 25 countries will be randomized 1:1 to receive either 400 mg Rinzimetostat daily plus Darolutamide or, at the physician's discretion, an androgen receptor inhibitor or Docetaxel. ORIC Pharmaceuticals presented preclinical data on rinzimetostat, an allosteric PRC2 inhibitor for prostate cancer, at the AACR meeting. Analyses of more than 1,100 samples showed that PRC2 activity occurs early and is elevated in over half of localized primary tumors. Rinzimetostat demonstrated antitumor activity in combination with darolutamide in models ranging from castration-sensitive to castration-resistant prostate cancer and retained efficacy in EZH1‑overexpressing cells, where mevrometostat and tazemetostat were less effective.
