Alector presents brain-shuttle pipeline and AL137 plan at Cantor health conference

Alector showcased its transferrin receptor–based brain-carrier platform and lead Alzheimer’s candidate AL137, which is slated to enter a Phase I study with subcutaneous dosing in April. Preclinical data show markedly higher drug concentrations in the brain without a significant drop in red blood cells; AL137 is intended to provide full effector function with reduced anemia risk. Alector share falls after failed phase‑3 study of latozinemab

Alector reports that the phase‑3 INFRONT‑3 study of latozinemab (AL001) in frontotemporal dementia due to a progranulin mutation did not meet its primary endpoint, prompting a sharp decline in the stock in after‑hours trading. Despite increases in plasma progranulin concentrations, secondary and exploratory endpoints showed no treatment‑related effects. Alector is halting the study extension and the continuation study with GSK, plans a workforce reduction of about 49%, and will focus on remaining programs such as Nivisnebart (AL101/GSK4527226) and BBB‑platform candidates AL137 and AL050.
