ORIC Pharmaceuticals starts global phase‑3 study Himalayas‑1 in metastatic castration‑resistant prostate cancer
ORIC Pharmaceuticals has launched the global phase‑3 Himalayas‑1 study for patients with metastatic castration‑resistant prostate cancer after treatment with Abiraterone. About 600 patients at more than 250 sites in 25 countries will be randomized 1:1 to receive either 400 mg Rinzimetostat daily plus Darolutamide or, at the physician's discretion, an androgen receptor inhibitor or Docetaxel. The primary endpoint is radiographic progression‑free survival; the most important secondary endpoint is overall survival. Bayer is supplying Darolutamide free of charge, and ORIC retains all worldwide development and commercialization rights. The dose selection is based on phase‑1 data and discussions with the FDA; the company has a market capitalization of $1.09 billion and, according to analysis, more cash than debt. » More on de.investing.com
ORIC Pharmaceuticals presents preclinical data on rinzimetostat in prostate cancer at AACR
ORIC Pharmaceuticals presented preclinical data on rinzimetostat, an allosteric PRC2 inhibitor for prostate cancer, at the AACR meeting. Analyses of more than 1,100 samples showed that PRC2 activity occurs early and is elevated in over half of localized primary tumors. Rinzimetostat demonstrated antitumor activity in combination with darolutamide in models ranging from castration-sensitive to castration-resistant prostate cancer and retained efficacy in EZH1‑overexpressing cells, where mevrometostat and tazemetostat were less effective. The company emphasizes that rinzimetostat can overcome acquired resistance mechanisms; ORIC has a market capitalization of about $1.09 billion, more cash than debt, and does not expect to be profitable this year. » More on de.investing.com
Oric Pharmaceuticals stock plunges after disappointing Rinzimetostat data
Oric Pharmaceuticals shares fell 41% on Wednesday and dipped further on Thursday after early study data were released for the prostate cancer candidate Rinzimetostat in combination with Darolutamid. In the dose-optimization study of 18 patients the confirmed PSA50 response rate was 33% and rPFS rates were 93%, 84% and 84% after three to five months, lagging behind comparable Mevrometostat data. Analyst reactions ranged from a 'peer-perform' rating to confirmed buy recommendations with price targets, while Oric plans to start the phase-3 Himalayas-1 study in H1 2026. » More on de.investing.com
